Multicenter phase 2 trial of first-line camrelizumab plus chemotherapy for NSCLC with brain metastases: final results of CAP-BRAIN

The phase 2 CAP-BRAIN trial at seven hospitals in China evaluated the combination of first-line camrelizumab plus pemetrexed and carboplatin for four cycles, followed by camrelizumab plus pemetrexed maintenance for up to 24 months in patients with metastatic non-squamous non-small cell lung cancer. Final results of the trial have now been published. At data cutoff, the median duration of follow-up was 46.7 months. The updated confirmed intracranial objective response rate was 57.5% (95% CI 40.9-73.0). Median intracranial PFS, overall PFS, and OS were 8.4 months (95% CI 6.2-not reached), 7.2 months (4.5-21.0), and 18.4 months (10.3-29.9), respectively. Treatment-related adverse events were reported in 91.1% of patients. There were no unexpected safety signals. The authors conclude that these results show promising benefitand manageable toxicity of first-line camrelizumab plus pemetrexed and carboplatin in non-squamous NSCLC patients with brain metastases.
