Multicenter phase 2 trial of FAK inhibition for meningiomas with somatic NF2 mutations
Dr. Priscilla Brastianos
Patients with progressive or recurrent meningiomas have limited therapeutic options once surgery and radiation fail. Focal adhesion kinase (FAK) inhibition has a synthetic lethal relationship with loss of neurofibromatosis 2 (NF2). Given the predominance of NF2
mutations in meningiomas, the phase 2 Alliance A071401 trial evaluated the FAK inhibitor GSK2256098 for recurrent or progressive grade 1-3 meningiomas. GSK2256098 was well tolerated and resulted in improved progression-free survival rate at 6 months compared with historical controls.
