De-escalated neoadjuvant chemotherapy strategies are need for HER2-positive breast cancer. The multicenter phase 2 NJMU-BC01 trial in China evaluated neoadjuvant de-escalated nab-paclitaxel combined with pyrotinib (small-molecule irreversible pan-HER receptor tyrokinase inhibitor) and trastuzumab in intrintrinsic HER2-enriched breast cancer. The study found promising efficacy of the regimen, with grade 3 or 4 treatment-related adverse events in 23% of patients, but no grade 5 TRAEs and no treatment discontinuation due to disease progression during the neoadjuvant treatment.
