Treatment options for recurrent or metastatic nasopharyngeal carcinoma (RM-NPC) with progression after PD-1 blockade are not clear. Immunotherapy combined with angiogenic therapy has been reported to have synergistic antitumor activity. A multicenter phase 2 trial in China evaluated the combination of camrelizumab plus famitinib for RM-NPC after failure or PD-1 inhibitor-containing regimens. The ORR was 33.3% (90% CI 15.6-55.4) and the DCR was 77.8% (56.1-92.0). The median time to response was 4.2 months, the median duration of response was 4.2 months (90% CI 3.0-NR), the median PFS was 7.2 months (4.4-13.3), and the median OS was not reached during median 16.7 months of follow-up. Treatment-related serious adverse events occurred in 33.3% of patients, with no deaths due to TRAEs. The authors conclude that camrelizumab plus famitinib had encouraging efficacy and tolerable safety in patients with RM-NPC after failure of frontline immunotherapy.
