AK112 is a novel bispecific antibody targeting PD-1 and VEGF. A phase 2 trial at 11 centers in China evaluated the combination of AK112 and platinum-doublet chemotherapy for advanced non-small cell lung cancer. The combination had promising activity as first-line therapy for aNSCLC without known driver mutations. The combination also had activity for aNSCLC with EGFR-functional mutation after failure of previous EGFR-TKI and aNSCLC after failure of systemic platinum-based chemotherapy and PD-(L)1 inhibitor therapy. The authors conclude that the combination of AK112 plus platinum-doublet represents a valuable new treatment option for aNSCLC patients.
