The clinical significance of adjuvant chemotherapy after resection of lung metastases from colorectal cancer (CRC) is not clear. A multicenter phase 2 trial in Japan evaluated adjuvant mFOLFOX6 in this setting. Patients received up to 12 adjuvant cycles of mFOLFOX6. The 5-year overall survival rate was 85.2% (95% CI 71.4-92.6) and the 5-year disease-free survival rate was 60.2% (44.9-72.4). Forty-one of 52 patients (78.8%) completed 12 cycles of mFOLFOX6. The most common grade ≥ 3 adverse events were neutropenia (50.0% of patients), fatigue (7.7%), and peripheral sensory neuropathy (7.7%). The authors conclude that adjuvant mFOLFOX6 is safe and may be effective after resection of lung metastases from CRC.
