Multicenter phase 2 study of upfront FOLFOXIRI-bevacizumaz with or without atezolizumab for metastatic colorectal cancer

In previous studies, immune checkpoint inhibitors had only limited activity for proficient mismatch repair (pMMR) or microsatellite stable (MSS) metastatic colorectal cancer. Both active combinition chemotherapy like FOLFOXIRI and bevacizumab seem to be able to increase the immunogenicity of pMMR or MSS tumors. The multicenter randomized phase 2 AtezoTRIBE trial in Italy evaluated addition of atezolizumab to first-line FOLFOXIRI plus bevacizumab for mCRC. Addition of atezolizumab to FOLFOXIRI-bevacizumab was safe and improved progression-free survival (HR 0.69; p=0.012).
