Multicenter phase 2 study of trastuzumab, pertuzumab, and docetaxel for advanced HER2 mutated NSCLC
Prof. Julien Mazières
HER2 exon 20 insertions and point mutations are oncogenic drivers found in 1%-2% of patients with non-small cell lung cancer. No targeted therapy is approved for this subset of patients. The multicenter phase 2 IFTC-1703 R2D2 trial in France evaluated the combination of two anti-HER2 antibodies and chemotherapy for pretreated HER2 mutated advanced NSCLC. Triple therapy with trastuzumab, pertuzumab, and docetaxel was feasible and effective, with ORR 29% and stable disease in 58% of patients. The median duration of response was 11 months (95% CI 2.9-14.0) and the median progression-free survival was 6.8 months (4.0-8.5).
