Multicenter phase 2 study of orelabrutinib for relapsed or refractory Waldenström’s macroglobulinemia

Orelabrutinib is a novel, small molecule, selective irreversible Bruton tyrosine kinase inhibitor. A multicenter phase 2 study in China evaluated orelabrutinib monotherapy in patients with relapsed or refractory Waldenström’s macroglobulinemia (R/R WM). The safety profile was manageable. The overall response rate was 89.4% and the major response rate was 80.9%. The PFS and OS rates at 12 months were 89.4% and 93.6%, respectively. The authors conclude that orelabrutinib had good efficacy and manageable safety in patients with R/R WM.
