The multicenter phase 2 CALM study evaluated intratumoral injections of the oncolytic coxsackievirus A21 (V937) for unresectable stage IIIC or IV melanoma. There were no treatment-related grade ≥ 3 adverse events. Regression of both injected and noninjected (eg, lung and lever) lesions occurred, indicating a systemic antitumor effect. The 6-month PFS rate was 38.6% (95% CI 26.0-52.4), the 12-month PFS rate was 32.9% (19.5-46.9), and the 12-month OS rate was 75.4% (62.1-84.7).
