A multicenter phase 2 study in China evaluated the combination of camrelizumab plus famitinib for recurrent or metastatic head and neck squamous cell carcinoma (R/M HNSCC) following progression after platinum-based chemotherapy. The study enrolled 20 patients, who received camrelizumab 200 mg every 3 weeks and famitinib 20 mg once daily. The primary endpoint was objective response rate. The objective response rate was 35.0% (95% CI 15.4-59.2) and the disease control rate was 55.0% (32.5-76.9); the median time to response was 2.23 months (2.0-8.3), the median duration of response was 8.0 months (2.2-NR), the median progression-free survival was 4.1 months (1.7-10.3), and the median overall survival was 8.4 months (3.0-17.0). Treatment-related adverse events of grade ≥ 3 included decreased white blood cell count (15%), hypertension (15%), pneumonia (15%), and tumor bleeding (15%). The authors conclude that camrelizumab plus famitinib demonstrated encouraging efficacy and manageable safety in patients with R/M HNSCC.
