Multicenter phase 2 study of adagrasib for previously treated NSCLC harboring a KRAS G12C mutation

Adagrasib irreversibly and selectively binds G12C-altered KRAS, locking it in its inactive state. A multicenter phase 2 study in the United States evaluated adagrasib for non-small cell lung cancer harboring a KRASG12C mutation in patients previously treated with platinum-based chemotherapy and anti-PD-(L)1 therapy. The ORR was 42.9%, the median duration of response was 8.5 months, the median progression-free survival was 6.5 months, and the median overall survival was 12.6 months. Treatment-related adverse events grade 1 or 2 occurred in 97.4% of patients and grade 3 or higher in 44.8% (including grade 5 in 2%) and resulted in drug discontinuation in 6.9%. The authors conclude that adagrasib showed clinical efficacy without new safety signals.
