Multicenter phase 2 basket trial of pembrolizumab plus vorinostat for recurrent or metastatic squamous cell carcinomas
Dr. Christophe Le Tourneau
The multicenter phase 2 basket trial PEVOsq in France evaluated the combination of pembrolizumab plus vorinostat for recurrent or metastatic SCC of various origins. The combination resulted in objective response rate of 26.2%, with ORR met in cohorts with SCC of cervix, anus, and vulva/vagina, but not of head and neck SCC or penile SCC. Median progression-free survival was 4,0 months (95% CI 2.6-4.3) and median overall survival was 11.1 months (9.2-17.4). The combination was tolerated with at least one TRAE in 91%, grade 3 TRAEs in 39%, and grade 4 TRAEs in 5.4%. The authors conclude that the combination of pembrolizumab and vorinostat showed promising antitumor activity in patients with R/M SCC with an overall manageable safety profile.
