Multicenter phase 1b trial of BLB01D1 for previously treated metastatic esophageal squamous cell carcinoma

BL-B01D1 is a first-in-class antibody-drug conjugate consisting of an EGFR-HER3 bispecific antibody bound to a topoisomerase I inhibitor payload via a cleavable linker. A multicenter phase 1 trial in China evaluated BL-B01D1 for previously treated metastatic esophageal squamous cell carcinoma. BL-B01DI had promising activity and manageable safety. The phase 2 dose was established at 2.5 mg/kg once every three weeks. A further phase 3 clinical trial has already been initiated.
