Multicenter phase 1b study of dostarlimab in doublet/triplet combinations for advanced cancer
Dr. Timothy Yap
Doublet combination therapies targeting immune checkpoints have shown promising efficacy in patients with advanced solid tumors, but it is unknown if rational triplet combinations will be well tolerated and associated with improved antitumor activity. The multicenter phase 1b study IOLite evaluated the PD-1 inhibitor dostarlimab in combination with the PARP inhibitor niraparib with or without bevacizumab, and carboplatin-paclitaxel with or without bevacizumab. The study found that dostarlimab was well tolerated in doublet and triplet regimens, with promising antitumor activity observed with all combinations. The disease control rates were higher with the triplet regimens than with the doublet regimens.
