Multicenter phase 1b-2 trial of second-line trastuzumab plus ramucirumab and paclitaxel for HER2-positive advanced G/GEJ cancer

Ramucirumab plus paclitaxel is the standard second-line treatment for patients with gastric or gastroesophageal junction (G/GEJ) cancer. A multicenter phase 1b-2 trial in South Korea evaluated the combination of trastuzumab plus ramucirumab and paclitaxel for HER2-positive advanced G/GEJ cancer after progression on trastuzumab-containing chemotherapy. The combination demonstrated appreciable efficacy and safety. HER2 expression was not correlated with the outcome in exploratory biomarker analysis. The authors conclude that continuation of trastuzumab in the second-line setting, in combination with paclitaxel-ramucirumab, is a promising strategy for advanced HER2-positive G/GEJ cancer.
