Approximately 3-4% of patients with non-small cell lung cancer have MET exon 14 skipping mutations. The phase 1b-2 GLORY study in China and Japan evaluated the selective, potent, oral MET inhibitor gumarontinib (formerly SCC244) monotherapy for locally advanced or metastatic METex14-positive NSCLC. Phase 2 results of the study have now been published. Gumarontinib had activity in both treatment naïve and pretreated patients, with ORR 71% (95% CI 55-83) and 60% (95% CI 42-76), respectively. Median progression-free survival was 11.7 months in treatment-naïve and 7.6 months in pretreated patients, and median overall survival was 17.3 months in pretreated patient and 16.2 months in pretreated patients. Treatment-related adverse events leading to discontinuation occurred in 8% of patients. The authors conclude that gumarontinib monotherapy had durable antitumor activity with manageable toxicity in patients with locally advanced or metastatic METex14-positive NSCLC.
