Multicenter phase 1b-2 trial of cadonilimab plus anlotinib as first-line treatment for advanced non-small cell lung cancer

Cadonilimab is a bispecific antibody simultaneously targeting PD-1 and CTLA-4. A multicenter phase 1b-2 trial in China evaluated the combination of cadonilimab and the VEGFR TIK anlotinib as first-line treatment for advanced non-small cell lung cancer (aNSCLC). The combination of cadonilimab 10 mg/kg every three weeks plus anlotinib 12 mg once daily resulted in treatment-related adverse events in 95.0% of patients, grade ≥ 3 TRAEs in 25.0%, and discontinuation due to TRAEs in one patient (5%). The confirmed objective response rate was 60.0%. The authors conclude that among patients with previously untreated aNSCLC, the chemotherapy-free combination of cadonilimab 10 mg/kg every three weeks and anlotinib 12 mg once daily had manageable safety and promising efficacy.
