Multicenter phase 1b-2 study of chiauranib with or without chemotherapy for platina-resistant or refractory ovarian cancer

Patients with recurrent ovarian cancer after platinum-based chemotherapy have a poor prognosis and limited treatment options. Chiauranib is a novel multi-targeted kinase inhibitor. A multicenter phase 1b-2 trial in China evaluated chiauranib monotherapy or in combination with etoposide or paclitaxel in patients with recurrent ovarian cancer after platinum chemotherapy. Chiauranib monotherapy resulted in objective response rate of 8.7% (panel A) and median progression-free survival 3.7 months (D), while the combination with etoposide resulted in ORR 40.9% (B) and median PFS 5.4 months (E), and the combination with paclitaxel resulted in ORR 52.4% (C) and median PFS 5.6 months (F). Adverse events were manageable. The authors conclude that this first study to evaluate chiauranib for recurrent ovarian cancer showed promising results. A phase 3 trial in ongoing.
