Datopotamab deruxtecan (Dato-DXd) is an antibody-drug conjugate consisting of an anti-TROP2 monoclonal antibody and a topoisomerase I payload. The multicenter phase 1 TROPION-PanTumor01 study evaluates Dato-DXd in patients with previously treated solid tumors. Results among patients with advanced or metastatic HR+/HER2- or triple-negative breast cancer have now been published. The objective response rate was 26.8% and 31.8% in the HR+/HER2- and TNBC groups, respectively. The median progression-free survival in the groups was 8.3 and 4.4 months, respectively. Grade 3 or higher treatment-emergent adverse events were seen in 41.5% and 52.3%, respectively. The authors conclude that among patients with heavily pretreated advanced HR+/HER2- or triple-negative breast cancer, Dato-DXd had promising clinical activity and a manageable safety profile.
