The phase 1-2 EMN09 study by the European Myeloma Network evaluated the combination of carfilzomib, bendamustine, and dexamethasone for relapsed or refractory multiple myeloma after at least two prior lines of therapy. The combination resulted in decrease in M protein in 56 of 63 patients. In ITT analysis 52% of patients had at least partial response, 32% had at least very good partial response, and 17% had complete response or near complete response. The clinical benefit rate was 93%. Median progression-free survival was 11.6 months, and median overall survival was 30.4 months. Grade 3 or higher AEs were lymphopenia (29% of patients), neutropenia (25%), thrombocytopenia (22%), pneumonia (13%), thromboembolic events (10%), cardiac events (8%), and hypertension (2%).
