Meta-analysis of treatment-related adverse events with HER2-targeted antibody-drug conjugates in clinical trials

Antibody-drug conjugates (ADCs) can improve oncological outcomes of cancer patients, but also are associated with treatment-related adverse events (TRAEs). Systematic review and meta-analysis of 39 studies involving 7688 patients across five cancer types investigated the incidence and grade of TRAEs of two HER2-targeted ADCs (T-DXd and T-DM1). The overall incidences of all TRAEs, high-grade AEs, serious AEs, and AEs leading to discontinuation were 98%, 49%, 19%, and 11%, respectively. The most common all-grade TRAEs werd nausea, fatigue, and decreased appetite, while the most common high-grade TRAEs were thrombocytopenia, anemia, and neutropenia. There were no differences in the mean incidences of TRAEs among different cancer types. T-DXd appeared to have higher mean incidences of TRAEs compared with T-DM1.
