Systematic review and meta-analysis of one randomized controlled trial and five retrospective cohort studies investigated the risk of thromboembolism with bortezomib-lenalidomide-dexamethasone (VRd) versus carfilzomib-lenalidomide-dexamethasone (KRd) for newly diagnosed multiple myeloma. The risk of venous thromboembolism was lower with VRd compared with KRd (6.16% versus 8.87%; OR 0.53; p=0.01) while there was no significant difference between the groups in risk of arterial thromboembolism (0.91% versus 1.16%; OR 1.01; p=0.99) (visual abstract).
