Lifileucel is an autologous centrally manufactured tumor-infiltrating lymphocyte product, that can be produced form harvested tumor specimens using a streamlined 22-day process. A multinational phase 2 study evaluated lifileucel for metastatic melanoma progressing after immune checkpoint inhibitors and BRAF ± MEK targeted agents. The ORR was 36% (95% CI 25-49), with two complete responses and 22 partial responses, while DCR was 80% (95% CI 69-89). Median duration of response was not reached after median 18.7 months follow-up. The safety profile was consistent with known adverse events associated with nonyeloablative lymphodepletion and interleukin-2.
