Intracranial efficacy and safety of lorlatinib for ALK-positive advanced NSCLC: post-hoc analysis of CROWN

Prof. Benjamin Solomon
Prof. Benjamin Solomon

Lorlatinib significantly improved progression-free survival versus crizotinib and showed robust intracranial activity in patients with previously untreated advanced ALK-positive NSCLC in the phase 3 CROWN trial. Efficacy outcomes in patients with and without baseline brain metastases and intracranial safety in CROWN have now been published. Lorlatinib improved PFS outcomes and reduced CNS progression versus crizotinib in patients with (panels A) or without (panels B) baseline brain metastases. Half of all CNS adverse events resolved without intervention or with lorlatinib dose modification.
