Interim overall survival results of abemaciclib plus nonsteroidal aromatase inhibitor for HR-positive, HER2-negative advanced breast cancer

The multinational phase 3 MONARCH 3 trial randomized patients with HR-positive, HER2-negative advanced breast cancer to a nonsteroidal aromatase inhibitor (NSAI) plus the CDK4/6 inhibitor abemaciclib or NSAI plus placebo. Interim overall survival analysis of the trial found that among all included patients and among patients with visceral disease, addition of abemaciclib to NSAI resulted in prolongation of median OS by more than 12 months; however according to the alpha spending procedure the increase of OS did not meet the threshold for formal statistical significance.
