Fulvestrant plus capivasertib for aromatase inhibitor-resistant ER-positive, HER2-negative metastatic breast cancer

The multicenter phase 2 FAKTION trial in the United Kingdom randomized patients with ER-positive, HER2-negative metastatic breast cancer after relapse or progression on an aromatase inhibitor to fulvestrant plus capivasertib or fulvestrant plus placebo. Updated results from the trial showed that addition of capivasertib to fulvestrant extended progression-fee and overall survival. Expanded biomarker testing suggested that capivasertib predominantly benefitted patients with P13K/AKT/PTEN pathway-altered tumors.
