Four-year follow-up of GAIA/CLL13: first-line venetoclax combinations versus chemoimmunotherapy for CLL in fit patients

The primary analysis of the multinational phase 3 GAIA/CLL13 trial showed that among fit patients with previously untreated CLL, venetoclax-obinutuzumab and venetoclax-obinutuzumab-ibrutinib improved progression-free survival (PFS) rates compared with chemoimmunotherapy. Up till now, no data on direct comparisons of different venetoclax-based combinations were available. Four-year follow-up of GAIA/CLL13 found that venetoclax-obinutuzumab and venetoclax-obinutuzumab-ibrutinib significantly extended PFS compared with both chemoimmunotherapy and venetoclax-rituximab. The triple combination had higher toxicities than the other regimens. The authors conclude that these results support the use of venetoclax-obinutuzumab in this patient population.
