Follow-up of ICARIA-MM study of isatuximab plus pomalidomide and low-dose dexamethasone for RRMM

Isatuximab is a monoclonal antibody that binds a specific epitope on the human CD38 receptor and has antitumor activity via multiple mechanisms of action. The multinational phase 3 ICARIA-MM study randomized patients with relapsed or refractory multiple myeloma after at least to previous lines of therapy, including lenalidomide and a proteasome inhibitor, to isatuximab plus pomalidomide and low-dose dexamethasone or pomalidomide and low-dose dexamethasone alone. With median follow-up of 35.3 months (IQR 33.5-37.4) the median overall survival was 24.6 months in the isatuximab group versus 17.7 months in the control group (HR 0.76; p=0.028). The authors conclude that isatuximab-pomalidomide-dexamethasone is a new standard of care for lenalidomide-refractory and proteasome inhibitor-refractory or relapsed multiple myeloma.
