Fixed-duration ibrutinib plus venetoclax versus chlorambucil-obinutuzumab as first-line treatment for CLL
Prof. Arnon Kater
The multinational phase 3 study GLOW compared the all-oral regimen fixed-duration ibrutinib plus venetoclax versus chlorambucil-obinutuzumab as first-line treatment for CLL/SLL in patients aged 65 years or older, or 18-64 years with CIRS score >6 or creatinine clearance <70 ml/min. The PFS was signficantly better with I+V than with Clb+O. I+V was also superior for the endpoints uMRD in bone marrow and peripheral blood, complete response, and time to subsequent therapy. The safety profile of I+V was consistent with treatmen in an elderly comorbid population.
