The multicenter phase 3 CameL trial in China compared 4-6 cycles of camrelizumab plus carboplatin-pemetrexed versus carboplatin-pemetrexed alone, followed by camrelizumab-pemetrexed or pemetrexed maintenance for previously untreated advanced non-squamous non-small cell lung cancer (ansqNSCLC). Crossover from chemotherapy to camrelizumab monotherapy was permitted after disease progression. Five-year outcomes of the trial have now been published. The HR for overall survival was 0.74 (p=0.0043) and was 0,62 (p<0.0001) after adjustment for crossover. Five-year OS rates were 31.2% with camrelizumab plus chemotherapy versus 19.3% with chemotherapy alone. Among patients who completed 2 years of camrelizumab, 5-year OS rate was 84.3%, and 5-year duration of respons rate was 46.5%. No new safety signals were observed. The authors conclude that camrelizumab plus carboplatin and pemetrexed continued to demonstrate long-term OS benefit over carboplatin and pemetrexed alone, with manageable toxicity. Patients who completed 2 years of camrelizumab had enduring response and impressive OS.
