Final overall survival analysis of phase 3 trial of pertuzumab retreatment for HER2-positive advanced breast cancer

In the primary analysis of the phase 3 PRECIOUS study, at 93 centers in Japan, pertuzumab retreatment combined with trastuzumab plus chemotherapy of physician’s choice (PTC; n=110) significantly improved investigator-assessed progression-free survival compared with trastuzumab plus physician’s choice chemotherapy (TC; n=109) among patiënten with HER2-positive locally advanced or metastatic breast cancer with progression after first- or second-line pertuzumab containing therapy. Final overall survival analysis of the study has now been published. The median follow-up was 25.8 months. The median overall survival was 36.2 months in the PTC group versus 26.5 months in the TC group (HR 0.73; 95% CI 0.55-0.97). The benefit of adding pertuzumab to TC was consistent across predefined subgroups. The authors conclude that these findings suggest that dual HER2 blockade with pertuzumab plus trastuzumab could contribute to improving overall survival in patients who have previously been treated with pertuzumab-containing regimens for HER2-positive LA/M BC.
