Final analysis of phase 2 GRIFFIN trial of addition of daratumumab to lenalidomide, bortezomib, and dexamethasone for NDMM in transplantation-eligible patients
Prof. Peter Voorhees
The multicenter randomized phase 2 GRIFFIN trial evaluated addition of daratumumab (D) to lenalidomide, bortezomib, and dexamethasone (RVd) for newly diagnose multiple myeloma (NDMM) in transplantation-eligible patients. Final analysis of the trial, at median 49.6 months follow-up, showed that addition of D to RVd improved the depth of response and the progression-free survival. Serious treatment-emergent adverse events occurred in 46% of patients in the D-RVd group and 52% of patients in the RVd group. The authors conclude that these results justify further evaluation in phase 3 studies.
