In the multinational phase 3 ALCYONE study, addition of daratumumab (D) to bortezomib, melphalan, and prednisone (VMP) significantly improved outomes of transplant-ineligible patients with newly diagnosed multiple myeloma (NDMM). Final analysis of ALCYONE, at a median follow-up of 86.7 months, has now been published. The median overall survival was 83.0 months with D-VMP versus 53.6 months with VMP (HR 0.65; p<0.0001). Most common grade 3 or 4 treatment-emergent adverse events were neutropenia (40% with D-VMP and 39% with VMP), thrombocytopenia (35% versus 38%), and anemia (18% versus 20%); grade 5 TEAEs were reported for five (1%) versus three (1%) patients. The authors conclude that with more than 7 years of follow-up, D-VMP continued to elicit clinical benefits over VMP in transplant-ineligible NDMM patients.
