Measuring CA-125 levels alone is widely used to diagnose progressive disease (PD) in patients receiving chemotherapy for platinum-sensitive recurrent ovarian cancer (PSROC). However, there are increasing concerns regarding its accuracy. Retrospective analysis of the phase 3 CALPYPSO trial (carboplatin plus pegylated liposomal doxorubicin versus carboplatin plus paclitaxel for PSROC) investigated concordance between CA-125 PD and RECIST PD. The analysis found that approximately 2 in 3 women who had RECIST PD did not have CA-125 PD. The authors conclude that monitoring CA-125 levels alone is not reliable for detecting PD.
