Crossover and rechallenge with pembrolizumab after recurrence in EORTC 1325/KEYNOTE-054

The phase 3 EORTC 1325/KEYNOTE-54 study randomized patients after complete resection of high-risk stage III melanoma for one year to pembrolizumab or placebo. In patients who crossed over to pembrolizumab after recurrence in the placebo group, pembrolizumab yielded a 39% ORR and median PFS of 8.5 months. Among patients who recurred more than six months after completion of treatment in the pembrolizumab group, pembrolizumab rechallenge resulted in median PFS of 4.1 months.
