CNS efficacy of osimertinib with or without chemotherapy in EGFR-mutated aNSCLC in the multinational phase 3 FLAURA2 trial

In a publication a month ago, the multinational phase 3 FLAURA2 trial showed that among patients with EGFR-mutated advanced non-small cell lung cancer (aNSCLC), addition of pemetrexed-platinum chemotherapy to first-line osimertinib was associated with significantly improved progression-free survival. Mandatory brain scans at baseline and progression allowed for systemic evaluation of CNS efficacy, which has now been published. In patients with baseline CNS metastases, the combination demonstrated a clinically meaningful improvement in CNS progression-free survival versus monotherapy (42% reduction in the risk of CNS progression or death); 59% versus 43% achieved complete response. CNS PFS benefit was also seed in patients without baseline CNS metastases.
