Associations of response rate and event-free survival with overall survival in newly diagnosed AML

Analysis of trial-level and patient-level data from eight randomized, controlled trials of intensive chemotherapy in patients with newly diagnosed AML examined the relationship between complet remission (CR) rate and event-free survival (EFS) with overall survival (OS). Trial –level analyses demonstrated a moderate association between hazard ratios for EFS and OS using a harmonized definition of EFS. Patient-level responder analyses demonstrated that patients who achieved a CR had better OS compared with CR without full count recovery and nonresponse. The authors conclude that use of CR rate and EFS as clinical trial end points could allow earlier evaluation of novel therapies for patients with newly diagnosed AML, and may correlate with OS.
