Association of TP53 gain-of-function versus non-GOF mutations with sidedness-dependent prognosis in mCRC

The prognostic significance of p53 mutations in colorectal cancer has not been established. A cohort study at Kaiser Permanente (Santa Clara, CA) investigated whether p53 mutations, when classified into gain-of-function (GOF) and non-gain-of-function (non-GOF) mutations could stratify the prognosis of right-sided versus left-sided metastatic CRC (RCC versus LCC). The study found that poorer survival of patients with metastatic RCC versus LCC appeared to be restricted to the subset with non-GOF p53 mutations, whereas GOF versus non-GOF p53 mutation was associated with poorer survival only among patients with LCC, and not among patients with RCC.
