Association of race with survival of prostate cancer patients treated with apalutamide: Pooled analysis of two phase 3 trials

Pooled analysis of the phase 3 TITAN and SPARTAN studies investigated the association of race with overall survival effect of the androgen receptor pathway inhibitor apalutamide among prostate cancer patients. TITAN and SPARTAN randomized patients to androgen deprivation therapy (ADT) with or without apalumide for metastatic hormone-sensitive and nonmetastatic castration-resistant prostate cancer, respectively. Among 2190 patients, 16.0% were Asian, 3.7% were Black, 67.4% were White, and 12.0% were Other. There was no significant heterogeneity of treatment effect of apalutamide on overall survival across racial groups (p=0.49). Among ADT plus apalutamide-treated patients, there was no association of race with overall survival: compared to White the HR was 0.77 (95% CI 0.56-1.06) for Asian, 0.82 (0.49-1.37) for Black, and 1.00 (0.75-1.34) for Other. The authors conclude that the analysis found no evidence of treatment effect of apalutamine on overall survival across patients of different races.
