A multicenter retrospective study in South Korea investigated the association of location of BRCA1 mutation or BRCA2 mutation with activity of frontline PARP inhibitor maintenance therapy in patients with advanced epithelial ovarian cancer. Among 380 patients, 242 (67.1%) harbored BRCA1 mutations and 137 (36.1%) harbored BRCA2 mutations (one patient harbored both). The study found a significant PFS benefit in patients with BRCA1/2 mutations, with pronounced benefits for those with mutation located in the DNA binding domain of BRCA1 and BRCA2; while there was no significant benefit of PARP inhibition for patients with mutations located in the C-terminal domain of BRCA1.
