Anti-CD19 CAR T-cell therapy for relapsed or refractory mantle cell lymphoma: five-year follow-up of ZUMA-2

The ZUMA-2 Cohort 1 study found high objective response rate (93%) and complete response rate (67%) among patients receiving brexucabtagene autoleucel (brexu-cel; autologous anti-CD19 CAR T-cell therapy) for relapsed or refractory mantle cell lymphoma (R/R MCL). Five-year outcomes of the study have now been published. Median follow-up for Cohort 1 (n=68) was 67.8 months (range 58.2-88.6) with a median duration of response of 36.5 months (n=60). Median overall survival was 46.5 months (95% CI 24.5-60.2) and was 60.2 months (42.8-NE) among 46 patients with complete response. The five-year cumulative incidence of relapse-related and non-relapse-related mortality was 40% and 22% in responders, respectively. No grade 5 CRS or ICANS, subsequent T-cell malignancies, or new safety signals were reported. The authors conclude that patients in ZUMA-2 continued to have durable responses after five years of follow-up.
