Anthracycline-free neoadjuvant chemotherapy with dual HER2 blockade for HER2-positive breast cancer
Prof. Gabe Sonke
The Dutch phase 3 study TRAIN-2 randomized women with HER2-positive stage II or III breast cancer to neoadjuvant treatment with 3 cycles of FEC followed by 6 cycles of paclitaxel-carboplatin or to 9 cycles of paclitaxel carboplatin. Both groups received trastuzumab and pertuzumab. Three-year follow-up of the study found similar EFS and OS between the groups, while the anthracycline receiving group had higher rates of febrile neutropenia, cardiotoxic effects, and secondary malignant neoplasms.
