The multinational phase 3 CATNON study randomized patients with newly diagnosed 1p/19q non-co-deleted anaplastic glioma 1:1:1:1 to radiotherapy, RT plus concurrent temozolomide, RT plus adjuvant TMZ, or RT plus concurrent and adjuvant TMZ. Concurrent TMZ did not improve overall survival (p=0.76). Adjuvant TMZ was associated with improved OS (median 82.3 months with adjuvant TMZ versus 46.9 months without adjuvant TMZ; HR 0.64; p<0.0001). Clinical benefit was dependent on IDH1 and IDH2 mutational status.
