Adding everolimus to first-line treatment of premenopausal advanced breast cancer after progression on SERMs

Prof. Binghe Xu

The effectiveness of the mTOR inihibitor everolimus in premenopausal women with HR-positive, HER2-negative advanced breast cancer after disease progression on selective estrogen receptor modulators (SERMs) is unknown. The multicenter phase 2 MIRACLE study in China evaluated addition of everolimus to letrozole in this setting. The median PFS was 19.4 months with everolimus plus letrozole versus 12.9 months with letrozole alone (HR 0.64; p=0.008). Among patients with progression on letrozole alone, crossover to everolimus plus letrozole resulted in median PFS of 5.5 months.
